In horses, TIMP-1 demonstrates multifaceted functions beyond its role as a metalloproteinase inhibitor. Extensive research has revealed its involvement in various physiological and pathophysiological processes. TIMP-1 is secreted by different cell types, including fibroblasts, synovial cells, and chondrocytes, and its expression can be regulated by factors such as mechanical stress, growth factors, and cytokines. TIMP-1 modulates the balance between extracellular matrix components, such as collagen, elastin, and proteoglycans, by inhibiting the activity of matrix metalloproteinases. This regulation is critical for maintaining tissue homeostasis, preventing excessive matrix degradation, and promoting proper tissue repair. Furthermore, TIMP-1 has been implicated in angiogenesis, the process of forming new blood vessels. It influences endothelial cell migration, proliferation, and tube formation, all of which are indispensable for the formation of functional vasculature during wound healing and tissue regeneration in horses. Studies have also highlighted the role of TIMP-1 in immune response modulation. It exhibits immunomodulatory effects by influencing leukocyte migration, regulating cytokine production, and modulating the activity of immune cells. This aspect of TIMP-1 function contributes to its involvement in various equine inflammatory diseases and immune-mediated disorders.
