S100-A9, also known as calgranulin B, belongs to the S100 protein family, a group of calcium-binding proteins celebrated for their involvement in various cellular processes, particularly those related to inflammation and immune responses. In horses, S100-A9 takes center stage in the innate immune system, predominantly produced by neutrophils and monocytes. The hallmark of S100-A9's functionality lies in its ability to form a heterodimeric complex with S100-A8, collectively referred to as S100A8/A9 or calprotectin. This complex is released by activated neutrophils and monocytes in response to inflammatory cues, thus serving as a sentinel of the innate immune response. Upon release into the extracellular environment, S100A8/A9 exerts potent pro-inflammatory effects, orchestrating the recruitment of immune cells to sites of infection or injury.
