α-defensin 5, or Def5, is a remarkable antimicrobial peptide found in the canine digestive system, specifically in the small intestine. It is a member of the α-defensin family, which comprises small, cationic peptides known for their potent antimicrobial properties. These peptides are pivotal components of the innate immune system, contributing significantly to the host's defense against invading pathogens. The synthesis and secretion of α-defensin 5 primarily occur within Paneth cells, specialized secretory cells located in the crypts of Lieberkühn in the small intestine. These cells are strategically positioned to defend the intestinal epithelium from harmful microorganisms. α-defensin 5 is part of this frontline defense mechanism, actively engaging and neutralizing a wide range of bacteria, viruses, and fungi that may enter the digestive tract. Structurally, α-defensin 5 possesses a unique arrangement of cysteine residues that form disulfide bonds. These bonds not only provide structural stability but also enhance its antimicrobial activity.
