N-terminal prohormone of brain natriuretic peptide (NT-proBNP) is a circulating biomarker that is derived from the precursor molecule proBNP, which is synthesized in the cardiac ventricles of animals, including bovines/cattle. ProBNP consists of a 108-amino acid polypeptide that is cleaved into two distinct fragments, N-terminal proBNP (NT-proBNP) and active brain natriuretic peptide (BNP). NT-proBNP is released into the bloodstream in response to cardiac stress or injury, as well as mechanical and neurohormonal stimuli. It is a marker of cardiac strain and dysfunction, reflecting increased wall tension in the ventricles. The concentration of NT-proBNP in the bloodstream reflects the degree of cardiac impairment, making it a valuable biomarker for monitoring cardiovascular health and predicting adverse events in bovines/cattle. NT-proBNP achieves its vasodilatory and natriuretic effects through the interaction of natriuretic peptide receptor A (NPRA) and NPRA-associated guanylate cyclase domains. The resulting increase in cyclic guanosine monophosphate (cGMP) leads to the relaxation of vascular smooth muscles and increased salt and water excretion. NT-proBNP has a relatively long half-life of approximately 1-2 hours, making it a useful diagnostic tool as it remains elevated for more extended periods than the active hormone BNP. It can be measured in serum or plasma using various immunoassays, including enzyme-linked immunosorbent assay (ELISA) and chemiluminescent immunoassay (CLIA). NT-proBNP levels are age and breed-specific, with healthy animals exhibiting lower levels than those affected by cardiovascular disease.
